In the lab I am working in, we are growing cells 3h, 37°, inducing them with IPTG when OD is nearly 0.6.

After 3h, they are harvested for purification.

Usually they take 80-90 minutes to reach 0.6, but last time I worked with an old plate and they took more than twice. At 3h, instead of having an OD between 1-2, they had 0.7-0.8, which is very little. I am concerned it will affect the protein purification and concentration.

May the slowing down be due to the plate 1 week old?
