I am doing experiments with MCF 10A cells, which were cultured in DMEM/F12 supplemented with 5% horse serum, 20ng/ml of EGF, 0.5µg/ml of hydrocortisone, 100ng/ml of cholera toxin, 10µg/ml of insulin and pen/strep. The DMEM/F12 media has 15mM of HEPES, 0.5 mM sodium pyruvate, and 1200 mg/L sodium bicarbonate. How do I supplement this media recipe for cell culture in ambient CO2 level? I have to run an experiment in a setup which does not have the 5% CO2 supply.
