After co-cultive is essential that the cellular machinery assimilates the T-DNA region ( usually this region also contain the genes implicated in resistence to herbicides). This step must be maintained for 1-2 days, since usually this medium content acetosyringone as inductor of T-DNA transfer and insertion during Agrobacterium infection. One time the T-DNA region is inserted into genome, the plant can produced the genes implicated in resistence to herbicides and not before.
The main purpose of resting stage is to recover as many independent transgenic events as possible. Unless the multiple transgenic events would fuse together making it difficult to recover the events separately (Hiei and Komari, 2008).
