I have a flow cytometry profile in which the proportion of cells in S phase decrease continuously after increasing time of drug treatment whereas the number of cells in G1 phase increases moderately and proportion of G2/M cells increase mildly. So there is a conflict as to this is a G1 phase arrest or G1 phase delay. Therefore I want to know how to differentiate between a cell cycle arrest and a delay.
There really no difference, other than cell cycle delay implies that the cell will come out of the checkpoint and re-enter the cell cycle. Cell cycle arrest implies permanent G0. Most likely you are seeing a transitory arrest at the Restriction point in late G1. You might want to look at p53 levels in the G1 fraction.
Moderate G1 arrest point out that mTOR signaling is affected. Try rapamycin as positive control to test this.
