I tried to measure the fluorescence intensity of pharmaceutical compound using (Multi-mode plate reader, Perkinelmer EnSight) instrument and the fluorescence intensity appeared as RFU not RFI and its value is very high (around 1 million).
So, i am asking about the difference between RFU and RFI ? and what about this high value ? and is it possible to use it for pharmaceuticals ?

Different companies (instruments) may report the fluorescence units (RFU or RFI) using different relative units. The units are arbitrary (which is why they are referred to as "relative"). You should not compare results obtained from two different instruments. Ideally, run samples on one instrument, at the same time, for comparison.
If you run identical standards (cal curves) on two different instruments you could probably develop a relative correction factor to use when comparing them for the same sample. Establish one instrument as the "standard" reference instrument with a value of '1', then determine the factor or ratio for the other.
