If I have to develop a Rotenone-induced Parkinson's model in the SH-SY5Y cell line, and further check the neuroprotective effect of my synthesized compound, does it really affect whether I treat my synthesized compound after or before rotenone treatment. My simple question is that the maximum number of papers suggest the pretreatment of a neuroprotective compound before treating with Rotenone for inducing PD in SH-SY5Y cells. What will happen if I treat Rotenone first before my synthesized compound?

Generally for plant extracts and nutraceuticals we go for pre-treatment models and for marketed or potent compounds we go for post-treatment models. If you have synthesised compounds that are potent antioxidant and anti-inflammatory I would suggest you to go for post treatment methods.
However from my work in SH-SY5Y cells I have differentiated the cells with Retinoic Acid (10uM) in reduced serum media for 7 days. Following induction with MPP+ (100uM - 4hrs) or Rotenone (100nM - 24hrs) we gave treatment and found good results.
