Has anyone tried co-culturing and SH-SY5Y cells with human astrocytes and differentiating ? I am planning to seed astrocytes, let them grow for 3 days and introduce SH-SY5Y cells into it. Ideally, adding Retinoic acid followed by BDNF is done to differentiate the SH-SY5Y cells into the neuronal phenotype as explained by many published protocols. I am wondering if this is feasible with astrocytes in the co-culture. Any experienced input will be really appreciated!

There are certain things to be optimised before working with co-culture of astrocytes and SHSY5Y neurons.

- Culture Conditions: Both cell types might have slightly different culture requirements in terms of media composition, seeding density, and incubation conditions. Optimizing these parameters is crucial for successful co-culture.
- Co-culture Method: There are different techniques for co-culturing cells, such as physical separation using Transwell inserts or direct co-plating on the same culture dish. Choosing the right method depends on the desired level of cell-cell interaction.
