What is the difference between Oligo(dT)12-18 Primer and Anchored Oligo dT? because the price variation seems too high in Invitrogen. And incase of Oligo (dT)12-18, they suggest 0.5ug for < 5ug of total RNA. How much should i use for 1ug of total RNA?
An anchored Oligo(dT) primer ends with a V (a mixture of A, C, and G). Such a primer is supposed to bind at exactly at the first base upstream of the poly(A) tail of the mRNA. To make it work specifically you may have to play with the annealing/RT temperature.

If you are doing it for the first time..go exactly by the protocol recommended by the kit you are using ...to avoid any problems right in the begining

if you are buying your primers from a company for a protocol they are always much more expensive than just getting a primer manufacturing company to make the primer to your design just like your pcr primers...you get more and they cost less
