I tried embedding it in low melting point agar but it peeled off the agar during sectionning. I tried cutting it without the agar since it is an adult brain but i disintegrated altogether. I am aiming for a saggital section of around 250um thickness. My vibratome doesn't have an O2 chamber i don't know if that is a problem but the tissue is sectionned within artificial CSF . Any suggestions please?

you can try to mount it with super glue/immediate adhesive glue on a plastic block and then you mount brain and block in your vibratome.
The brain and the cutted slices needs then to float in artificial liquor.

Yes i tried it but because it's too fragile it keep detaching from the glue clock as well while being cut.
Some times lower Temperature (the buffer) can be helpful, also new sharp blade is a good option. Indeed agar and brain position (the way you locate and cut them) is very important.
