hello, Im having trouble in extracting information from my uv-vis data. The sample the I analyzed is protein corona. Before that, I already characterize 40 nm of gold nanoparticle and obtain its absorbance is 0.9 and at wavelength 526.50 nm. I read some journal the absorbance of protein corona will decrease, but the wavelength remain the same.
From my UV-vis data, my protein corona graph has no peak and no known wavelength, it just a flat graph. Can I know others opinions?

Blue shift means increase in frequency, i.e., decrease in wavelength. On the other-hand, red shift denotes the reverse case (As, in visible spectra blue is at highest energy end, and red is at the lowest energy end). In the UV-Vis absorption spectra you have denoted X-axis in nm (wavelength), so blue shift means the absorption peak should shift in the left side.
From the curve you have provided, it seems one absorption peak is at a wavelength ~526 nm, and the other two are below 300 nm.
I Think that nanoparticles have been conjugated with the proteins and all the surface was coated, for that the light do not 'see' the correct surface of Au-NPs. verify with FTIR if the proteins were grafted to the surface of NPs

Bathochromic shift: In spectroscopy, the position shift of a peak or signal to longer wavelength (lower energy). Also called a red shift. A hypsochromic shift is the shift of a peak or signal to shorter wavelength(higher energy). Also called a blue shift.

Could you specify structure of your ligand and metal-complex, without structure you can just imaging what happens after coordination. It might be changing of the relaxation pathways from IL to MLCT, which is much more higher in energy
Bathochromic shift is the Redshift ( shift towards longer wavelength)
Hypsochromic shift is the Blueshift ( shift towards shorter wavelength)
