Concentration of folate binding protein (sample) is 2,5 - 3 mg/ml after dialysis. I am going to use 12 mL DEAE cellulose 52 (by journal), my lecturer ask me about capacity matrix but i don't know exactly because there is no information in the package. so then I need to know the amount of sample required to be loaded on the column to get good resolution?

According to this web page, the protein binding capacity of DE52, in terms of bovine serum albumin (molecular weight 66 kDa) is 550-700 mg per gram of dry resin and the swollen volume is 6.5 ml/g. Therefore, your 12 ml column is made of about 2 g of resin, with a binding capacity of about 1 g of BSA, which is about 15 micromoles.
