Native Gymnopilus sp. and Xylaria polymorpha mycelium is grown in PDA from spores and tissue. Transfers of the healthy portion of hyphal growth were taken several times for each species.
One PDA dish with Gymnopilus sp. transfer is fully colonized and some PDA dishes are only starting to colonize. However, I need to store the PDA dishes for several weeks. What is the best methodology to do so? Moreover, when I put the dishes back in the incubator at optimum mycelium growing temperature, will there be any negative impact on the growth rate?
Can prepare Slate culture. Due to the small surface area of the slate, it can be store as permanent culture.

If you want to store the plates for several weeks, you can left them in the incubator at 25 C or in the refrigerator at 8 C and then, if you want to work with them you must refresh them by preparing sub culture from them and incubate these subcultures in incubator again for about 4-7 days.
If you want to store the living cultures for a long time, you can apply more than one method. For example, you can use glycerol and distilled water 10% or 30% in small glass tubes (2ml) for storing living culture. The tubes may store under lab temperature or under deep freezing (-80 C).
Also, you can maintain the living cultures of fungi by liquid Nitrogen under -196 C. In this method, all biosynthesis processes of the fungus are stopped. So the fungi can be preserved for a long time about many years.
There is a simple method for preserving fungal living culture. It represented by using mineral oil and glycerol which added to the slant contains the fungal culture with a deep of 1 cm. In this method the living culture can be preserved for about 2-3 years.

Lyophilization is another important method to preserve living culture. In this method, the fungal culture was immersed with 1,2 Polyurethane and alcohol, then it dried at 50-60 C and prepared a suspension and put in glass tubes. This method will allow to preserve many samples in small space without any contamination. The living culture can be stored for a period reaches 20 years.
