I want to evaluate the antimicrobial activity of several plant extracts (extracted by supercritical CO2 extraction method - e.g.sage,oregano,) against Salmonella carnosus. The extracts are oily and rarely watersoluble. I need recommendations for suitable dilution reagent. Also I would like to know which kind of test for determining the MIC you would suggest? My lab wants to try broth macro dilution method...
Some workers have emulsified with nonionic surfactant (Tween) and others solubilized in DMSO or ethanol. Please be aware that for publication in most significant journals you'll need not only analysis of oils to the significant constituents but also some novelty as there are numerous reports of MIC testing of sage and other essential oils

You may use Tween 80 , DMSO, ethanol, and DMF any one solvents in antimicrobial testing for oily compounds They worked well at around 1% (w/w) in culture media or PBS.
Important thing to follow is to use a vehicle control to make sure the solvent itself has no antimicrobial effect on the microorganisms.
