What may be the best way to assay diacylglycerol (DAG) extracted from rat liver? Some references say about ELISA kit and some says about the conventional assay for DAG is lipid extraction DAG separation by LC and then MS. Any suggestion is greatly appreciated.
I'd say LC-MS-APCI is the simplest way to identify/quantify DAGs after extraction with a non polar solvent

The best way to assay DAG is to perform TLC, then scoop the silica portion containing DAG and then perform GC.
After Folch extraction and removing solvent you could derivitize to TMS ethers and run on a high temperature GC column.This seperates FFA,monoglycerides ,diglycerides and triglycerides but not by stereospecifity. The others suggestions of LC/MS is better if this is the question you need answering
