Has anybody positive experience with RNA isolation from forest fungi ? I was trying to isolate RNA from Boletus and Suillus fruiting bodies. RNA was isolated. After RT and cDNA PCR I have got positive results with internal parts of my gene of interest. But when I tried to amplify whole gene I was not successful. mRNA was probably degradated. Is there any effective and sensitive way of isolation RNA ? I have tried different kits and also isolation by Phenol-Chloroform-Isoamyl alcohol.
