I attached the pics of protocol which i follow in my lab for RNA isolation from Candida and it works well with everybody in my laboratory (with the kit used is from Qiagen). I can see you mentioned "growing on plate" you and grow them in same medium in broth and after overnight growth you can perform RNA extraction. You can refer qiagen protocol too. You can use CTAB based method (manual protocol) or kit as well. As rightly suggested by Dhirend, the best way is to growth the cultures in liquid medium overnight (or even more, depending on species, especially filamentous fungi are slower to growth than yeasts). After that, to obtain best results, I suggest to filter dry and lyophilize the mycelia. Regarding kit, I tried both Qiagen Rneasy and the Spectrum Plant Total RNA Kit from SIGMA ALDRICH and I can assure that the Spectrum Plant Total RNA Kit can give you higher yield and higher quality.
