Can anyone please suggest the best protocol for NBT staining (for suproxide radicals) in tomato leaves. Could you also suggest how, after staining, I can efficiently remove chlorophyll without damaging leaves and heat burning.
I tried this protocol....leaves got stained with this...but i unable remove chlorophyll..chlorophyll is partially get removed...i thank you for your positive reply...
What you need to do is remove the already 'green' ethanol for a new one, until you don't see green parts. It takes some days, but at the end, leaves turn white.
After staining, you can put leaf into boiling ethanol (96%) to remove chlorophyll without damaging leaf. The detailed method is attached here. I had tried this protocol and gotten very good result.
