Invertase activity on sucrose may be monitored by determining the apparition of glucose. Glucose may be monitored continuously using for instance a coupling assay with hexokinase in presence of ATP and G6Pdehydrogenase in presence of NAD+. The stoechiometric generation of NADH is followed at 340nm. The other main coupled assay to determine glucose is glucose oxydase coupled to peroxydase in presence of a chromogenic substrate.
Sucrose inversion can also be monitored by optical rotation, using a polarimeter, or by thin-layer chromatography on silica gel 60 plates, using multiple ascents in 90% acetonitrile (aq).
