I'm making functional nanocomposites with iron oxide nanoparticles for MRI contrast agent. While making the sample for injection, figured out that we need to calculated exact amount of Fe ions in unit volume of sample.

You can use Spectroscopic method like ICP-AES but digestion of your sample should be done carefully. Also, you should take care of the number of times you are diluting the sample before analysis, as it required before final sample preparation. Most importantly you should know the volume/amount of sample you are actually loading in the instrument for precise calculation of the iron content.
Secondly, you can used the colorimetric method also for iron detection called as ferrozine based assay, there is a difference of some +/- 0.2 in the final concentration of Iron.
Both the techniques are equivalent
But before doing these two, you can estimate the Iron content using Phenanthroline method too.
Hence you can compare the data from all the three.
