H. pylori is a fastidious organism capable of growing only under a narrow set of conditions. It must be cultured on nutrient-rich agars such as Columbia blood agar base supplemented with whole blood from any one of a variety of large mammals such as horse, ox, or sheep. Reduced oxygen tension is crucial; therefore, a closed-container system capable of generating microaerophilic conditions is usually employed. Many laboratories have had success growing H. pylori on plates using a tissue culture incubator set at 37°C and either 5 or 10% CO2 with a tray of distilled water set in the bottom of the incubator to maintain humidity. Depending on the isolate, inoculum dose, and freshness and moisture of the plates, growth is generally observed between two and five days. However, some laboratories have been unable to achieve H. pylori growth using this method. Therefore, the most widely reliable method remains a closed-container system.
