I am conducting research on oral immune tolerance induction with FVIII protein. My initial goal is to check the DC population increment in payer's patches. I have read out different works of literature and found CD11chiCD11b+CD8α−, CD11chiCD11b−CD8α+, and CD11chiCD11b−CD8α− subtypes are recommended. I am expecting suggestions from an expert.

You can use fluorescence tagged Abs against those markers which only present on the DC cells. If you want to looking for migration of DC cells in that case you can use two markers one present on DC as well as other cells present on the payer's patches cells and second one only present on DC cells in that case you monitor cells migration into in payer's patches. Hope it will help you to design your experiments.
