I want to extract the total proteins in liver of a mouse for analyzing several proteins, including phosphorated proteins. What is the best lysis buffer? It would be better if there were reagents protecting phosphorated proteins in the lysis buffer.
For sure you would need to use a lysis buffer containing both protease and phosphatase inhibitors. There are many in the market, even cocktail containing most of them.

Hi Jianbo, just like Antonio J Ruiz Alcaraz said that the lysis buffer which named RIPA buffer and you should add protease(in our lab 1 pill used in 10ml RIPA lysis buffer) and phosphatase inhibitors (in our lab 1~2 pills used in 10ml RIPA lysis buffer) . and then mixing enough and always keep in ice box before use... hope can help you~
