Lieishmania donovanni is a blood flagellated protozoan parasite that may infect man and cause visceral lieshmaniasis. Phlebotomus sand fly is the insect vector.
M-199, RPMI-1640 and DMEM are the most frequently used media for in vitro passaging of L. donovani parasite. If you are directly taking parasites from patients, then Novy-MacNeal-Nicolle (NNN) medium is used for the culture of Leishmania parasite. The incomplete medium should be prepared with HEPES, NaHCO3, L-Glutamine and antibiotic cocktail (without amphotericin B). Finally, make complete media with the addition of 10% FBS to incomplete media (before use).
I agree with Sheetal. A good culture medium we use for the parasites of the Leishmaniinae group is very similar . It is M199 (Sigma−Aldrich) supplemented with 2 µg/ml hemin (Jena Bioscience, Jena, Germany), 10% heat-inactivated fetal bovine serum, 2 µg/ml biopterin, 100 units/ml of penicillin, and 100 µg/ml of streptomycin at 23 ºC.

The best culture media used for of the Leishmaniinae promastigotes in Schneider’s Drosophila medium (Sigma-Aldrich) at 22°C and sub-cultured as promastigotes in M 199 (Sigma-Aldrich) both supplemented with 100 U/ml penicillin, 100 μg/ml streptomycin, and 10% FCS (Sigma-Aldrich).
