1) Before in vivo test in rat LAD ligation model I want to test cardiomyocyte VEGF lipofection with cationic vectors we've elaborated. I want to test them on ventricular cardiomyocytes. Does anybody knows the best cardiomyocyte cell line to do this and where could I find it?
2) I already have mouse endothelial cardiac cell to make co-culture with cardiomyocytes. Do I need to use same species cardiomyocytes ?
HL-1 cells are a mouse atrial myocyte lineage but I think they are only available from the Claycomb laboratory. Primary neonatal mouse ventricular mycoytes are fairly easy for most labs to isolate, or you can buy them from Lonza. Neonatal myocytes are modestly transfectable with lipofection type reagents, although usually adenovirus is used for high efficiency. There is a decent JOVE article on the primary isolation J. Vis.
