I have difficulty to chose the right vector to anayze my interest gene tissue expression in Rice. I have two candidate vectors, such as pCambia1391z and pMpGWB104. Both vectors contain C-terminal GUS fusion. In pCambia1391z, although 35S promoter directed to the hygromycin resistance gene, it very close to the insert region (35S-> Hygromycine before insert region). Therefore, I feel that it might still affect promoter activity. In pMpGWB104, 35S-> Hygromycin is after GUS sequence. However, 35S direction is toward the GUS and I am not sure if it still affects GUS activity (antisense and sense direction). And also, this vector was not tested in rice. There also other alternatives, such as citrine and mCherry. I would really appreciate your suggestions and experinces if you did promoter analatysis in Rice.
