Transfection works very well in vitro because most in vitro transfection involves cells undergoing division. While in the in vivo most recepient cells are non dividing or slow dividing cells where rate of transfcetion is low. This is evident from the low rate of transfection in the in vitro conditions of primary cells,which exhibit low rate of cell division.

Keeping the above in mind what are the approaches that facilitate a decent rate of transfection?

Device mediated transfections such as gene gun mediated delivery and ultrasonic delivery are some approachjes which provide some low level of transfection. PEI mediated delivery is another approach which has been applied in gene delivery by various routes. Liposomal methods work excellently in in vitro conditions however I do not know if they work well in in vivo gene delivery.
