I have a mutation in cystathionine beta synthase (CBS) gene cloned in bacterial pET28 vector and I would like to clone it in a yeast vector. I already amplified the gene by PCR, did PCR purification, restriction enzyme digestion for both the gene and yeast plasmid. I would like to ask what is the best yeast transformation protocol to proceed with this experiment?
