what do you mean by extracting the DNA from butter? do you mean the DNA of microorganisms or the DNA of somatic cells?

I suggest to do centrifuge at ambient temperature, to separate the fatty material from the proteins, then I suggest to use dneasy blood and tissue kit, the main problem is, there may be different species of microorganisms.

if there is many species of microorganisms, you may try to create a primer with specific regions to isolate specific kinds of microorganisms
