From soil we can isolate the bacetria/fungi by traditional serial dilution techniques. If anyone working on biodegaradtion than let me know about the screening parameters for microbes with respect to biodegradation??? May be your active research experience is different and useful for me???
You have to be more specific regarding biodegradation of xenobiotics, cellulosics, ligno-cellulosics, oil-spill etc. What type of biodegradation you wish to look for in the soil microbes? Usually direct isolation methods give to visualization of degraded substrate(s) because of clear zone formation around cfu/ fungal growth.
Dear Professor, Thank-you very much for your reply. I am seeking for pesticide degrading microbes from soil.?Although I am doing the routine practices like cfu based study in presence of pesticide/or optical density in minimal media.. besides this i am looking for any other suggestion for the same to select the potential strains.

Best approach is to add the desired pesticide at a lower concentration in a selective medium, and allow the growth of soil microbes (Diluted soil microbes added) in it at 37 C for 3-4 days. Subculture and enrich these organisms gradually in higher concentration of pesticide (added to broth) and you are likely to get some microbes capable of pesticide degradation. This always have been a hot topic of research!

Another way how to select microorganisms for bioremediation could be performance of respirometric tests using pollutant as a substrate. The results for every microorganism are easy to compare.
I would tell that in real world the bioremediation is performed mostly by microbial consortia rather than a single microbial strain. So be careful about this fact.

I agree with Vit. The mere development of colonies on selective media cannot come off as bio-degradation. The best it demonstrates is bio-utilization. Respirometry and carbon dioxide evolution methods are better methods for biological degradation measurement. You could also measure the dehydrogenase activity of the organism. It correlates very strongly with respirometric data. All the best!

Soil is a dynamic home for many organisms. I could suggest you some points which might allow you to screen larger community of organisms of interest.
* Collect various soil samples. For example, top layer soil, middle layer soil, soil from the contaminated site etc.,. This will allow you to screen wide spectra of organisms.
* Use different nutrients rich media (like TSB, TGY), do not restrict to single media for isolation. You can also give a try with minimal media.
* Soil can be treated with the pesticide for certain time duration before serial dilutions (Different weight of soil can also be tested).
* In the selection of the BEST isolate for remediation purpose, you can test with individual and combination of 2 or 3 pesticides.
