1 Inducing 8-week old Wistar rats (male 500g and female 250g) with 65 mg/kg STZ IV to create a diabetic peripheral neuropathy model. To cause these animal to develop a neuropathy in about 17 weeks, what would be an ideal target blood glucose level to maintain?
2) Immediately after the STZ procedure, BGLs spike to the 200-400 range, and after hours are extremely low (40 mg/dL) and may need to be maintained using glucose IP. Would anyone recommend switching to a multiple-injection (spread out over several days) induction of DM1 by STZ for this study?
Many litterateurs suggested the dose for a single high dose in mice ranges from 100 to 200 mg·kg−1 (Srinivasan and Ramarao, 2007; Dekel et al., 2009), depending on the mouse strain (Hayashi et al., 2006), and in rats 35–65 mg·kg−1 (Srinivasan and Ramarao, 2007). This leads to a rapid ablation of the beta cells and hyperglycaemia.
