What is the air humidity inside of a Petri dish (94x16) with fresh PDA medium, inoculated with 0,5 ml of spore suspension, at 25 degrees Celsius, during a 32 hours incubation? It depends on the incubator humidity? I need saturated atmosphere for 32 hours. Thank you very much in advance!

It will depend on a number of factors, eg do your petridishes have vents, the humidity of the incubator, is the temperature stable, are your plates directly on a heating element (or lamp) or under a blower that switches on or of during the day. The first day the PDA will provide enough water to keep very high humidity but in most incubators the plates start drying after 2 days. You can do a few things to ensure high humidity, eg put them plates in a box with some wat tissue paper at the bottom or close the plates with micropore (which leaves O2 through but keeps water in) or parafilm (closes both water nad air out). You can't use parafilm if you need a lot of respiration. I also grow them upside down to prevent condensation droplets running over the plate.

hi... i have another question. i have a leaf inoculated with fungus in an empty petri dish. how do i induce humidity in the petri dish? should i put in a wet filter paper or something? anything better? pls help and thank u
If the incubator is fitted with a humidifier then it will solve the purpose of maintaining the humidity provided the petridish would not be tightly closed.
If not to give moist condition you can wet filter paper / damp cotton / water agar.
