What is the sham dose?
How should we use its data for mtt assay analysis?
I heard that if my drug solvent is PBS should take a dose of it and add to my cells like control, is it correct?

If you're asking about using a control in your experiment, you should always include a control if it is possible. In that way, you can compare to an experimental unit that has not been exposed to the factor that you're investigating, but is treated the same in every other aspect. So, yes, you treat this control using only the solvent, at the same volume as all the other treatments.

So mtt assay should have two control one is cells with the complete medium without any treatment. And another is sham that is cells with solvent treatment and medium. Hope I understand correctly. Also, it should have wells consist of the blank for removing background noise. If my solvent is water or alcohol should I use sham?
You're welcome. Yes, it doesn't matter what your solvent is, you just want to do everything the same to have a comparison of 'cells treated only with solvent'.
