Initially i have done bleaching of cotton Fibre which is then followed by acid hydrolysis by using 100ml of sulphuric acid (50%).Form of cotton doesnot change' it still seems like raw cotton. How can i recognize that cellulose nanocrystals has formed? Does the internal morphology has been changed?

You may need to give more detail information on how to treated it, condition, temperature, time ...
After reaction time, your need to washing, separation, and collect the CNC from the synthesis source. You can observe the morphology of cotton -CNC by SEM, TEM, AFM ... I think the procedure and condition that you did may not success, that is why form of cotton doesn't changed. Or in the washing stage you did removed all the CNC?
You should read some detail methods on CNC preparation then it will work.

Please can any share the procedure for making cellulose Nanocrystal from microcrystalline cellulose or any can share his/her work.
Did you treat your fibers mechanically after hydrolysis? Once you mechanically treat them (e.g. ultrasonication), then you can centrifuge your sample. The supernatant will contain your nanocellulose fibers. Atomic force microscopy is used for evaluating the aspect of the fibers produced. But as Hai Le Van mentioned, you need to share your process conditions.

I'm adding a chart that I've used in my lectures summarizing the size ranges for the various divisions of cellulose fibers. I hope this helps you to determine what you've produced. AFM and TEM can measure nanofibers. SEM will be able to measure microfibers and larger. The majority of processes can be summarized as follows;
- 1) Chemical, Mechanical, Chemical/Mechanical treatment of fibers
- 2) Washing (dialysis)
- 3) Surface chemical analysis
- 4) Mechanical defiberization (ultrasonication, microfluidization)
- 5) Centrifugation (supernatant contains the nanofibers, larger particles will be in sediment).
- 6) Concentration (rotary evaporator)
- 7) Analysis
A literature review of your selected process should provide you with enough detail to replicate treatment conditions. A quick test to see if you have produced nanofibers is to take your supernatant and dry it at 50C in an oven. If a clear film forms, you have nanofibers. Usually the supernatant will appear cloudy at low surface charge. I'll attach a photograph showing what the supernatant looks like at constant concentration as charge increases.
