I know the estimate degree of gene expression (EDGE) method is a comprehensive approach that is suitable for data with or without replicate, and for both single group and multiple group tests.
But I don't know what and when do we have to use the EDGE?

EDGE has different meanings: Empirical Analysis of Digital Gene Expression (for differential gene expression) or Estimated Degree of Gene Expression, to my knowledge, for estimating the level of RNA expression in Prokaryotes.
I don't know what you plan to do. Gene expression analysis is generally conducted not to estimate the absolute level of gene expression but rather to identify differentially expressed genes between two or more biological conditions. And there is no way to escape biological replicates, since the rationale is based on comparison of mean average and standard error.
