I'm trying to conceive a set of experiments to test the orientation of the nucleosomes relative to each other in a nucleosomal array that has assumed the 30 nm conformation. Any thoughts or previous attempts? This is the most recent review I know of - "Higher-order structures of chromatin: the elusive 30 nm fiber." Anyone have a more recent paper?
From what I can tell, there is a growing consensus that the 30nm fiber does not really exist. See "Human mitotic chromosomes consist predominantly of irregularly folded nucleosome fibres without a 30-nm chromatin structure" by Nishino et al. EMBO Journal 2012, and the accompanying article "Human mitotic chromosome structure: what happened to the 30-nm fibre?" by Hansen in the same issue.
Of course, not everybody thinks so - see "Chromatin organization — The 30 nm fiber" by Grigoryev and Woodcock in Exp. Cell Res. 2012.
Personally, I don't see the need for a 30nm fiber in interphase nuclei, at least - a more de-compacted and flexible model seems to be more realistic.
Apologies, I just looked at your profile and it looks like you've published with a few of the authors I just cited, so you must know those papers. Excuse me if I've misunderstood your question.
Justin, Thank you for your answer. I have been away from science for the past year and a half so even though I have published with Jeff, I was not aware of this recent review/research. I must look like an idiot for not being up on these but hey, how else do you learn :)
