I am interested in performing SDS PAGE of proteins isolated from drosophila larva muscle. I am using the following lysis buffer: 150 mM NaCl, 50 mM HEPES, 1% Triton-X, 10% glycerol, 1.5 mM MgCl2 and 1.0 mM EGTA) plus protease inhibitors. I have also used RIPA buffer in the past which seems to be the common lysis buffer. But, I am looking for a milder lysis buffer to lyse the tissue. Does anyone have any experience using milder lysis buffer to lyse larval musles?
