Some papers report performing a "STD self intensity" test to check the aggregation of compounds in STD NMR. Can anyone please elaborate how such a test is performed?
The test is done by running an STD experiment on the ligand alone, without the presence of a macromolecule, such as a binding protein. This is to make sure that all the signal you see in the actual STD experiment with the ligand and macromolecule is in fact from the interaction of the two, and not from the ligand alone.
The experimental parameters should be identical to those used for the actual interaction study.
