I am planning a study to test the effectiveness of a cancer therapy on reducing tumor volume in a 4T1 mouse model. In addition, I would like to see the effect of my treatment on the metastatic spread of the tumors into the lungs. I recently came across a paper where they were able to analyze this with Q-RT-PCR which allows them to detect small metastases that cannot be seen with the traditional counting method by looking at the upregulation of genes that are specific to the cancer cell. In their study, they used human HER2. I dont think this would work for me since 4T1 cells are triple negative. Can anyone else suggest another marker which I could use for this? We have previously demonstrated that orthotopic transplantation of 4T1 cells showed lung metastasis, in which RNA-binding protein ESRP1 (epithelial splicing regulatory protein 1) alters the alternative splicing machinery of CD44, p120, and FGFR2 genes.

Hi, As the matrix metaloproteinase enzymes (MMPs) are involved in both angiogenesis and metastasis of solid tumor, so evaluation of their expression level may be helpful in your study! MMPs family contain 13 type of enzyme but MMP2 and MMP9 are more related to metastasis and are more important.

Thank you for your thoughtful responses. I have also been looking into some options and have seen some evidence that Ki67 may be a good candidate. Does anyone have any insights into this?
