phytochelatins is metal peptide bonds occur due to heavy metal stress, or it is a defence mechanism of plants. my queries is for best possible lab method to determine this pc bond in plant? i need with good reference protocol.

Please try electrophoretic profiling method for thiol-rich phytochelatins and metallothioneins. You can find the detail methods with Phytochem Anal. (2004) 15(3):175-83.
I did that experiment with following process.
1. seqeunce homology search of known PC or MT / metal binding sequnce
(heavy metal tolerant species such as brassica napus)
2. cloning the candidate genes and transform into yeast
3. Comparing heavy metal resistance between WT and transformants by spot test
