Hello everyone, I try to purify a High molecular weight protein. But i have a big problem because it forms aggregates. I try to add glycerol (5-10%) or increasing the NaCl concentration of 0,15M to 0,5M. But when i did appear two new peaks in the size exclusión column ( Ve: 9,8 and 11,35 ml, the column is a Superose 6) rather than one it is the result when i did without these additives ( Without the additives i obtained one protein peak in Ve= 10,5ml). Why do these peaks appear? What happend in my protein purification?
I obtained other results where i saw the peak which Ve is 9,8 ml becomes the other ( Ve=11,5 ml). What is it happening? Is there any relation between the protein concentration and glycerol?
