I am trying to analyse the distribution of effector memory and central memory CD4 T cells in flow cytometry from my synovial fluid sample.I am getting CD62L+CCR7+ subpopulation which are central memory and CD62L-CCR7- as effector memory however a good percentage of my population are 62L+CCR7-.Can anyone please suggest me what does this 62L+CCR7- population represent.
Loss of CCR7 could indicate that central memory cells are late differentiated.

The expression pattens of CD62L and CCR7 in T-cell subsets may be as follows.

CCR7 + + + – – –
If there has been no updates to the information (I am not at all sure), then I suppose the CD62L+CCR7- cells could be an undocumented subpopulation of T cells. I could even suspect that the so-called transitional memory T cells may be better characterized to clarify the expression patterns of markers such as CD27, CD28, CD62L and CCR7. So I guess it would be highly informative if you could further investigate the subset.
By the way, from your text you may be analyzing T cells in the patients with Rheumatoid Arthritis? I might be also important to clarify whether you are interested in T cells in a physiological or pathological setting.

actually CD62L+CCR7+ are not Central memory, expression of CD62L+CCR7+ means that they are in the first step of differentiation, only naïve T cell express CD62L+ and CCR7+ you can call this gate naïve like population inside this regarding to expression of CD62L and CD95 you can distinguish between Tscm and Tte and Tcm and Tem.
Thanks@ Mojtaba Shekarkar. But we generally use CD45RA and CD45RO for gating naive and memory cells that is CD45RA+ CD45RO- are naive one... According to your answer it means we need to use CD95 for gating central memory thanks.... We will try it.
