For MTT assay, I was supposed to treat the cancer cells mcf7 (96-well plate ) for 3 hours with solution of chemotherapic drug made by “free-FBS RPMI medium”. But mistakenly, I made solution with complete medium. Any difference may cause in results?

yes complete medium cause difference in result because fbs reaction with mtt substance

The difference in results could occur due to several reasons:
1) cells usually do not like medium without FBS and the viability in that case could be lower;
2) in the presence of 10% FBS the compound solubility in medium could be different (its penetration, and thus toxicity to the cells could be slightly different).
Low concentrations of FBS are recommended. Always use the same concentration in all assays. I agree with Vilma Petrikaite.
