For different donors, we observed the upregulated expression of immune checkpoint ligands on monocytes when they were cultured with only CD8 T cells (both antigen-specific and non-specific CD8 T cells).

Expression was not observed when monocytes were cultured (without T cells) either alone or with a cancer cell line. Moreover, expression was not observed for 3-cell culture of monocytes, cancer cell line and non-specific T cells, but greatly upregulated when the 3-cell culture instead involved antigen-specific T cells.

(Media used contains 100IU/mL of IL-2 to 'keep T cells happy'.) Appreciate any input on this strange phenomenon.
