I did PMBC cytokine (TH1/TH2) stimulation assay with POKEWEED, CONCANAVALIN-A, and PHYTOHEAMAGLUTININ . Pokeweed and PHA showed same level of cytokine secretion but CON-A had low level of secretion in all the cytokines. What could cause concanavalin-A to secret low level of cytokine compared to PHA and pokeweed.

Which species .....?, because conA is sometimes not working, horse and Elephant T cells are not stimulated by ConA. type IV...
Pure T cells are also not stimulated with conA ....you need to add APC to your culture.
ConA is crosslinking proteins of APC and T cell . and binds specifically α-D-mannosyl and α-D-glucosyl residues of receptor on cells.

PHA is better for human PBMCs. However these mitigens are not the same thing and various cells respond based on interaction and cross-linking.
