I am working on Shiga toxin producing E.coli (STEC). I have presumptive isolates and i want to determine their sero groups, that is O157 and non O157 as well as their virulent genes, stx, eae, hlya. I want to use multiplex PCR because i have so many samples and i want to save time. i have challenge optimizing my conditions because of varied annealing temperatures. 

If you have worked on this before, could you tell me the optimum conditions you used. I also need advise.
