During my study I have found in Combretum ovalifolium of in vitro formation of aerial roots at the region of node along with shoot. Are any references to this available?
@ Sir Gaber Abogadallah., Its without any auxin., Accidently it is found during nodal culture of this plant.
@ Leonardo Dias., I have used TDZ, 2-ip, Kinetin and BAP. Formation of roots occurs without any auxin, and interesting thing is that formation of aerial root along with shoot from nodal region., the question is that at the same time axillary meristem have dual function in this species. first bud break--- than--- shoot ----- formation of aerial root from the same region (aerial root: formation/initiation of roots above the medium).
First of all thank you Sir@:Gaber Abogadallah, In the prsent study formation of arial roots along with axillary proliferation from same node here is tthe image of result, firstly shoot formation............followed by rooting..........in the same medium but interesting thing is that formation of root is not from basal region of the cut end of the node., here root is formed from nodal region. My doubt is that "Is same meristem have dual function in vitro"
If auxin is used then why root not originate from cut end, why from base of node i.e. my question. I have used auxin inhibitor also TIBA to block endogenous auxin transport. but result is same
Try use ethylene scavengers or membranes that provide more gas exchange in in vitro atmosphere. Polyamines and brassinosteroids maybe have involved in this response.
It is obvious that nodal region has meristem that can produce both axillary bud and adventitious roots. Physiological age of the explant may be the reason to form the roots at the nodes and it is common in some plants when you use the nodes/ mature nodes/nodes from mature plants. If you want to avoid the root formation start the nodal explants from the in vitro shoots, that may stop the root formation.
