I am performing explants on marine invertebrates and out of all of the organs, the striated muscle specifically has yellowing media with turbidity. The bottom of the tissue culture plate seems to be coated in something but I am unsure of the possible contaminant. Has anyone observed this under the microscope? I have attached an image at 20X where you can see the plate covered. If you could provide some literature I can reference that would be great. Thank you.
I have no specific experience with marine organisms, but are you sure it is contaminated? Yellowing and turbidity could also be caused by the high energy consumption of the cultured cells, we see this routinely during differentiation of human iPS.
Are there any major differences between the cultures of the different explants that could account for a contamination?
In case this is a contamination it would be best to take a medium sample for analysis, maybe you have access to a facility that can help you with that.
