I have been using an Anion Exchange column for 1.5 years now. I recently encountered a low pressure issue when the outlet of column is attached to the detector.
For my assay desirable pressure is 385-420 psi , however i am getting low pressure of 71-141 psi.
System pressure (without attaching the column) is zero, when column is attached it show a pressure of 212 psi but on attaching the detector inlet to column outlet a drop in pressure is observed ie there is no backpressure. As a result of this a steady baseline is not obtained.
pH of the buffers, temperature conditions, resin elution, flow lines have been checked.
How can I troubleshoot this?

The pressure decreases once you connect the column to the detector? Prior to connecting it, the pressure is OK? That sounds strange.
What type of AEX column are you using, Sephadex? Could it be that the column has collapsed?

Yes, prior to connecting it to the detector the pressure is as expected. It indeed is strange and is happening for the first time.
I am using a column from GE, it appears to be fine. I did not see any resin or anything coming out of the column. I have been monitoring it continously.
The only thing that has been changed is ferrule which is used to tighten the detector inlet into the column outlet. We were previously using a metal ferrule which is now replaced with a plastic one. Can it cause a pressure difference?

It's weird that your pressure drops significantly on connecting the column to the inlet of the detector. Did you try connecting a different column to the same instrument to rule out the possibility that there is no issue with the flow lines, connections and the instrument itself?

Yes, other columns were connected to the detector as the same instrument is used for different applications. But those columns did not show any any pressure fluctuations or difference.
Do let me know if there is any way of troubleshooting this issue.

Looks like you're column has gone bad. Probably issues with column packing.
