In measuring ERK1/2 levels in a homogenous sample, what can contribute to increased ERK1/2 levels apart from loading different amounts of total protein or the phosphorylated state of ERK1/2.
So my question is in short what can cause an increase in ERK1/2 levels from baseline?
Activation of map kinases by growth factors are the main cause of increased erk 1 and 2 increase. But I suppose your question goes beyond this. Please can you be more specific?
Tomas Koltai thank you for your answer. Iam doing a wb for a sample cultured with TNF alpha which is known to increase ERK1/2 phosphorylation. However, i also noticed an increase in ERK1/2 (not phospho ERK). The result shows that there is an increase in ERK1/2 phosphorylation but also an increase in ERK1/2 levels from baseline. I don't know how to explain this result. I am looking for a resource that talks about Mapks transcriptional activators but all i find is Mapks phosphorylation activators.
We have seen changes in total ERK1/2 immunoreactivity in heart associated with remodelling. At this point, I have stayed away from it as changes in total ERK1/2 immunoreactivity could involve changes at any or all of the following: transcription, mRNA stability, translation, protein stability degradation. Each of these processes are subject to regulation.
