Has anyone seen this immunohistochemistry artifact, and do you know what causes it?
The artifact appears as patches of chromogen throughout the tissue sections. I have seen this artifact in multiple tissues, especially mouse pancreas, salivary gland, and mammary tumor. It looks like there may be lifting of the tissue off the slide in these regions, allowing antibody/chromogen to be trapped. I have been using positively charged glass slides to ensure the tissue adheres to the slide. Still, this is a repeatable artifact and I am not sure how to prevent it. Any advice would be appreciated.
